OPT-302 + aflibercept is a combination therapy for neovascular (wet) age-related macular degeneration (AMD). OPT-302, also known as sozinibercept, is a recombinant fusion protein that acts as a "VEGF-C/D trap," inhibiting vascular endothelial growth factor C and D. Aflibercept is an anti-VEGF-A agent that also blocks VEGF-B and placental growth factor (PIGF). The rationale for the combination is to achieve dual inhibition of VEGF-A (by aflibercept) and VEGF-C/D (by sozinibercept), targeting multiple pathways involved in pathological angiogenesis in wet AMD. The combination was evaluated in the COAST Phase 3 trial but did not meet its primary endpoint of superiority over aflibercept monotherapy in improving best corrected visual acuity at 52 weeks[2][3][4][6][9].
