OR2805 + cemiplimab

OR2805 + cemiplimab is an investigational combination therapy for advanced solid tumors. OR2805 is a fully human monoclonal IgG1 antibody that selectively binds to CD163, an immunosuppressive scavenger receptor highly expressed on M2 tumor-associated macrophages (TAMs). By targeting CD163, OR2805 reprograms M2-like macrophages toward a pro-inflammatory, M1-like phenotype, thereby relieving immune suppression and enhancing T-cell activation within the tumor microenvironment. Cemiplimab, marketed as Libtayo, is a monoclonal IgG4 antibody targeting PD-1 on T cells, blocking interaction with its ligands to restore anti-tumor immune responses. The combination aims to synergistically enhance anti-tumor activity: OR2805 reduces macrophage-mediated immunosuppression while cemiplimab releases inhibition on T cells via PD-1 blockade. This regimen is currently being evaluated in phase 1/2 clinical trials for subjects with advanced or metastatic solid tumors, including non-small cell lung cancer (NSCLC), melanoma, liposarcoma, leiomyosarcoma, and squamous cell carcinoma of the head and neck[1][2][3][5][6].
