Oregovomab + paclitaxel is an investigational combination therapy being studied primarily for the treatment of advanced epithelial ovarian cancer. Oregovomab is a murine IgGκ1 monoclonal antibody that specifically targets the tumor-associated antigen CA125 (also known as MUC16), which is commonly overexpressed in ovarian cancer. By binding to CA125, oregovomab forms immune complexes that are taken up by antigen-presenting cells, leading to enhanced processing and presentation of CA125-derived peptides to T cells. This process aims to break immune tolerance and stimulate a robust anti-tumor immune response against CA125-expressing tumor cells[4][6]. Paclitaxel is a well-established chemotherapeutic agent that stabilizes microtubules and inhibits cell division, leading to apoptosis in rapidly dividing cancer cells. The combination of oregovomab with standard chemotherapy (paclitaxel plus carboplatin) has shown promising results in phase 2 trials, with significant improvements in progression-free survival (PFS) and overall survival (OS) compared to chemotherapy alone[5]. A confirmatory phase 3 trial (FLORA-5/GOG3035) is ongoing to further evaluate efficacy and safety in newly diagnosed patients with advanced ovarian cancer following optimal debulking surgery[1][4][8].
