Oregovomab + poly ICLC is an investigational **combination immunotherapy** comprised of **oregovomab**, a murine monoclonal antibody targeting the tumor biomarker CA-125, and **poly ICLC** (polyinosinic-polycytidylic acid stabilized with poly-L-lysine and carboxymethylcellulose), an immunostimulatory double-stranded RNA analog that acts as a viral mimic and potent immune booster. Oregovomab acts as an immunization strategy, binding to circulating CA-125 and forming immune complexes that stimulate both humoral and cellular anti-tumor immune responses. Poly ICLC activates innate immunity through pattern recognition receptors, notably TLR3 and MDA5, leading to production of cytokines and chemokines, activation and infiltration of effector T cells, and modulation of the tumor microenvironment. The combination aims to synergistically enhance anti-tumor immunity by inducing both robust antigen-specific and innate immune activation, with primary development in ovarian cancer and other immunotherapy-resistant cancers[5][6][9].
