Drug intelligence / Profile preview
"Orforglipron + insulin glargine" is an investigational combination regimen consisting of orforglipron (LY3502970), an oral, non-peptide, small-molecule glucagon-like peptide-1 (GLP-1) receptor agonist, and insulin glargine, a long-acting basal insulin analogue. Developed by Eli Lilly, this combination is evaluated for the treatment of type 2 diabetes in patients with inadequate glycemic control on basal insulin. Orforglipron acts as a partial agonist of the GLP-1 receptor, stimulating glucose-dependent insulin secretion, slowing gastric emptying, and reducing appetite, while insulin glargine provides a steady basal level of insulin to regulate glucose metabolism. In the Phase 3 ACHIEVE-5 trial (NCT06109311), adding once-daily oral orforglipron to titrated insulin glargine significantly improved HbA1c levels and reduced body weight compared to placebo, without increasing the risk of hypoglycemia.
