Overview

ORIC-944 + abiraterone acetate is an investigational **combination therapy** consisting of ORIC-944, a potent and selective **allosteric inhibitor of the polycomb repressive complex 2 (PRC2)** via the embryonic ectoderm development (EED) subunit, and **abiraterone acetate**, an androgen biosynthesis inhibitor targeting **androgen receptor (AR) signaling**. The rationale for the combination is based on targeting both epigenetic reprogramming (via PRC2/EED inhibition) and androgen pathway blockade, with the objective of treating patients with metastatic castration-resistant prostate cancer (mCRPC) who have progressed on prior AR pathway inhibitors. ORIC-944 is being developed by ORIC Pharmaceuticals, with early-phase studies exploring its safety and antitumor activity in combination with multiple AR pathway inhibitors, including apalutamide, darolutamide, enzalutamide, and abiraterone acetate. The combination with abiraterone acetate may be conducted in future studies, as listed in ongoing clinical trials[3][1].
