Overview

ORIC-944 + apalutamide is an investigational combination therapy under clinical evaluation for metastatic castration-resistant prostate cancer (mCRPC). ORIC-944 is a potent, highly selective, allosteric, orally bioavailable small molecule inhibitor of the polycomb repressive complex 2 (PRC2), acting via the embryonic ectoderm development (EED) subunit. Apalutamide is a well-established androgen receptor (AR) inhibitor. This combination is designed to overcome resistance to AR pathway inhibition by simultaneously inhibiting PRC2-driven epigenetic mechanisms and directly blocking AR signaling. Early-phase clinical trials have demonstrated promising PSA response rates and a safety profile compatible with long-term dosing[1][2][5].
