Overview

A combination therapy consisting of **ORIC-944**, a potent and selective allosteric inhibitor of the polycomb repressive complex 2 (PRC2) via the embryonic ectoderm development (EED) subunit, and **darolutamide**, an androgen receptor inhibitor marketed as Nubeqa. This combination is under investigation for the treatment of metastatic castration-resistant prostate cancer (mCRPC), particularly for patients previously treated with androgen receptor pathway inhibitors and up to one prior chemotherapy. Early clinical trials demonstrate substantial clinical activity (high PSA50 and PSA90 response rates) and a favorable safety profile, predominantly mild to moderate gastrointestinal side effects.[1][2][3][4][5]
