Overview

ABBV-CLS-484 (osunprotafib) + pembrolizumab is a combination therapy being investigated for the treatment of various solid tumors. ABBV-CLS-484 is a dual PTPN2/PTPN1 phosphatase inhibitor that enhances anti-tumor immunity through a unique dual mechanism of action. It targets tumor cells to suppress their growth while also promoting the activation of multiple immune cell types to increase their anti-tumor activity. The combination with pembrolizumab, a PD-1 targeting agent, is designed to potentially enhance efficacy compared to either agent alone. Preclinical data has shown that PTPN2/1 inhibitors have improved efficacy when combined with PD-1-targeting agents in multiple tumor models.
