OT-A201 + IMiDs is a combination therapy being developed by Onward Therapeutics for the treatment of advanced hematological malignancies. The regimen consists of OT-A201, a first-in-class humanized bispecific antibody targeting two immune checkpoints, and an immunomodulatory imide drug (IMiD) such as lenalidomide or pomalidomide. OT-A201, licensed from Biomunex Pharmaceuticals, utilizes the proprietary BiXAb technology to enhance anti-tumor effects through dual checkpoint inhibition and antibody-dependent cellular cytotoxicity (ADCC). The IMiD component acts as a molecular glue, binding to the cereblon (CRBN) E3 ubiquitin ligase complex to induce the degradation of transcription factors like Ikaros (IKZF1) and Aiolos (IKZF3), thereby stimulating T-cell and NK-cell activity. This combination is currently in Phase 1 clinical evaluation (NCT05009355) to assess safety and preliminary efficacy in patients with relapsed or refractory hematological malignancies.
