OT-A201 + lenalidomide is an investigational **combination therapy** comprised of OT-A201, a first-in-class humanized bispecific antibody targeting two immune checkpoints, and lenalidomide, an approved immunomodulatory small molecule (IMiD) with anti-cancer and immune-enhancing properties. OT-A201 binds two different immune checkpoint antigens and possesses a functional Fc domain to facilitate antibody-dependent cell-mediated cytotoxicity (ADCC). Lenalidomide modulates the immune system and exerts anti-tumor activity by binding to cereblon in the cullin ring E3 ubiquitin ligase complex, inducing degradation of key transcription factors, inhibiting pro-inflammatory cytokines, and enhancing NK and T cell cytotoxicity. The combination is being evaluated in a Phase 1 clinical trial for relapsed/refractory hematologic malignancies and solid tumors[1][3][5][9].
