Overview

This is a multi-agent chemotherapy and targeted therapy combination consisting of four drugs: - **Oxaliplatin** (a platinum-based chemotherapeutic agent that forms DNA crosslinks, inhibiting DNA replication and transcription) - **Capecitabine** (an oral prodrug converted to 5-fluorouracil, which inhibits thymidylate synthase and disrupts DNA synthesis) - **Gemcitabine** (a nucleoside analog that inhibits DNA synthesis by incorporation into DNA and inhibition of ribonucleotide reductase) - **Cetuximab** (a chimeric monoclonal antibody targeting the epidermal growth factor receptor [EGFR], blocking ligand binding and downstream signaling involved in cell proliferation). This regimen has been investigated in phase II clinical trials for advanced gastrointestinal cancers, including locally advanced pancreatic cancer and biliary tract carcinoma. The combination aims to maximize cytotoxic effects on tumor cells through multiple mechanisms—DNA damage, inhibition of nucleotide synthesis, and blockade of EGFR-mediated signaling pathways[1][2][5][6].
