Overview
Oxaliplatin + cisplatin + 5-fluorouracil is a combination chemotherapy regimen composed of three cytotoxic agents frequently used in the treatment of various solid tumors, particularly gastrointestinal cancers. Oxaliplatin and cisplatin are platinum-based compounds that exert their antitumor effects by forming DNA crosslinks, thereby inhibiting DNA replication and transcription and leading to apoptosis in rapidly dividing cells[6][8]. 5-Fluorouracil is a pyrimidine analog that inhibits thymidylate synthase, disrupting DNA synthesis and function[3][8]. This combination may be used in advanced or metastatic colorectal cancer as well as other malignancies such as gastric or esophageal cancer. The regimen leverages the complementary mechanisms of action to enhance cytotoxicity against tumor cells while attempting to overcome resistance seen with single-agent therapy[1][2][4].
