This investigational combination therapy regimen is being evaluated in the IPOXA clinical trial (NCT02866903), sponsored by Hospices Civils de Lyon. It is designed specifically for patients with peritoneal carcinosis arising from colorectal cancer. The regimen is unique in its delivery method, combining localized intraperitoneal (IP) administration of oxaliplatin with systemic (intravenous) FOLFIRI chemotherapy and bevacizumab. Oxaliplatin is a platinum-based cytotoxic agent that induces DNA damage through the formation of crosslinks. FOLFIRI is a standard-of-care chemotherapy cocktail consisting of folinic acid (leucovorin), 5-fluorouracil (5-FU), and irinotecan, which target DNA synthesis and repair. Bevacizumab is a biologic agent that inhibits angiogenesis by binding to vascular endothelial growth factor (VEGF). The Phase I/II study aims to determine if the high local concentration of oxaliplatin in the peritoneal cavity, combined with systemic therapy, improves outcomes for patients with difficult-to-resect peritoneal disease.
