Overview

This is a combination chemotherapy regimen consisting of three cytotoxic agents: oxaliplatin, gemcitabine, and vinorelbine. Each drug has a distinct mechanism of action targeting cancer cell proliferation. Oxaliplatin is a platinum-based compound that forms DNA crosslinks, inhibiting DNA replication and transcription. Gemcitabine is a nucleoside analog that inhibits DNA synthesis by incorporating into DNA strands and blocking chain elongation. Vinorelbine is a vinca alkaloid that disrupts microtubule formation during mitosis, leading to cell cycle arrest in metaphase. This combination has been investigated primarily for the treatment of advanced or metastatic cancers such as non-small cell lung cancer (NSCLC) and bladder cancer, particularly in patients unfit for cisplatin-based regimens or after failure of prior therapies[1][3]. Clinical studies have shown some activity with manageable toxicity; however, no significant efficacy advantage over two-drug regimens was observed[3]. Further clinical development beyond early-phase trials has not been widely pursued.
