Oxaliplatin + levofolinate calcium + 5-fluorouracil + bevacizumab is a multi-drug combination chemotherapy regimen, commonly referred to as mFOLFOX6 plus bevacizumab, used primarily for the treatment of unresectable, advanced, or recurrent colorectal cancer (mCRC). The regimen consists of the mFOLFOX6 backbone—comprising oxaliplatin (a platinum-based alkylating agent that induces DNA crosslinking), 5-fluorouracil (an antimetabolite that inhibits thymidylate synthase), and levofolinate calcium (a folate derivative that potentiates the activity of 5-fluorouracil)—combined with bevacizumab, a humanized monoclonal antibody that targets vascular endothelial growth factor A (VEGF-A) to inhibit tumor angiogenesis. This combination therapy is designed to simultaneously disrupt DNA synthesis and the tumor's blood supply. Takeda has been a significant sponsor in clinical trials, such as the Phase III PARADIGM study, which evaluated this regimen as a first-line treatment in patients with RAS wild-type metastatic colorectal cancer.
