Overview

This is an experimental **combination regimen** consisting of four agents: oxaliplatin (a platinum-based chemotherapeutic), raltitrexed (a thymidylate synthase inhibitor), TQB2868 (a bifunctional fusion protein targeting PD-1 and TGF-β), and ramucirumab (a monoclonal antibody targeting VEGFR2). This regimen targets malignant tumors through multiple mechanisms: traditional cytotoxic chemotherapy (oxaliplatin, raltitrexed), immune checkpoint inhibition and TGF-β pathway modulation (TQB2868), and anti-angiogenic therapy (ramucirumab). TQB2868 is under clinical development for indications such as metastatic pancreatic cancer and is designed to simultaneously block PD-1 and neutralize TGF-β in the tumor microenvironment, potentially restoring T cell activity and enhancing anti-tumor immunity[1][3][5]. Ramucirumab is FDA-approved for several cancers, including gastric, lung, and colorectal carcinoma, by inhibiting tumor angiogenesis through VEGFR2 blockade[2][4][6][8]. Oxaliplatin and raltitrexed are standard cytotoxics used primarily in gastrointestinal malignancies.
