Overview

The SOX regimen is a combination chemotherapy consisting of oxaliplatin, a third-generation platinum-based chemotherapy agent, and S-1 (TS-1), an oral fluoropyrimidine. Oxaliplatin exerts its cytotoxic effect by forming intra- and inter-strand DNA crosslinks, which inhibit DNA replication and transcription. S-1 is a multi-component drug containing tegafur (a prodrug of 5-fluorouracil), gimeracil (a dihydropyrimidine dehydrogenase inhibitor that increases 5-FU concentration), and oteracil (which reduces gastrointestinal toxicity). This combination has been evaluated in the Phase II TORCH trial (KCSG-BR07-03) led by Seoul National University Hospital for patients with recurrent or metastatic breast cancer previously treated with anthracyclines and taxanes.
