This digital medicine formulation consists of a combination of oxycodone (5 mg) and acetaminophen (325 mg) co-encapsulated with an FDA-approved ingestible sensor. Developed by the SWOG Cancer Research Network using Proteus Digital Health technology, the system is designed to monitor medication adherence in real-time. When the capsule is ingested, the sensor is activated by gastric fluid and transmits a signal to a wearable patch, which then relays the data to a mobile application and a healthcare provider portal. This technology is being evaluated in feasibility studies (such as SWOG S1615) to optimize pain management and monitor opioid utilization patterns in patients with metastatic cancer and uncontrolled pain. Oxycodone acts as a mu-opioid receptor agonist, while acetaminophen provides non-opioid analgesia, likely through the inhibition of prostaglandin synthesis in the central nervous system.
