Oxycodone + ziprasidone is an experimental combination of two FDA-approved drugs, oxycodone (an opioid analgesic) and ziprasidone (an atypical antipsychotic). This combination has been studied in pilot clinical trials to evaluate whether co-administration can maintain the analgesic efficacy of oxycodone while potentially reducing its abuse liability by blocking dopamine D2 receptors, which are implicated in opioid-induced euphoria. Oxycodone acts primarily as a mu-opioid receptor agonist to provide pain relief, while ziprasidone functions as a dopamine D2 receptor antagonist and also blocks serotonin 5-HT2A receptors. The rationale for this combination is that antagonism of the mesolimbic dopamine system by ziprasidone may limit the rewarding effects of opioids without compromising their pain-relieving properties. Early studies indicate that combining these agents does not significantly alter analgesia or increase adverse effects compared to oxycodone alone[2][3][4].
