**P22CYP-PpIX-PEG(EST)** is a preclinical multifunctional biocatalytic nanoparticle for **estrogen receptor-positive breast cancer**. It comprises a bacteriophage P22 virus-like-particle nanocage encapsulating cytochrome P450 activity, surface-conjugated protoporphyrin IX for UV365 nm-triggered photodynamic generation of reactive oxygen species, and a PEG-linked estradiol-derived ligand to promote estrogen receptor-mediated uptake by ER-positive tumor cells. In vitro, the platform increased intracellular CYP activity and enhanced tamoxifen sensitivity through combined enzyme-mediated prodrug activation and photodynamic cytotoxicity.
