p24CE + p55gag is a therapeutic DNA vaccine regimen developed by the University of California, San Francisco (UCSF) for the treatment of HIV infection. The regimen consists of two primary components: a p24CE DNA vaccine, which encodes seven highly conserved elements (CE) of the HIV-1 p24gag protein to focus the immune response on regions critical for viral fitness, and a p55gag DNA vaccine expressing the full-length HIV-1 Gag protein. These plasmids are typically co-administered with an IL-12 plasmid adjuvant via intramuscular electroporation. The vaccine is designed to elicit robust T-cell and humoral immune responses against structurally important regions of the virus that cannot easily mutate. It is currently being investigated in Phase 1/2 clinical trials as part of a combinatorial 'shock and kill' strategy aimed at achieving long-term HIV remission in individuals on antiretroviral therapy.
