The P2X3 antagonist being developed by Guangzhou Nanxin Pharmaceutical (also known as Nucien) is a small molecule drug candidate targeting the P2X3 receptor for the treatment of chronic cough, including refractory and unexplained chronic cough (RCC/UCC). P2X3 receptors are ATP-gated ion channels located on the sensory vagal fibers (C-fibers) in the airways. In patients with chronic cough, these receptors are thought to be hypersensitized by ATP released during airway inflammation or irritation, leading to an excessive cough reflex. By acting as an antagonist, this molecule blocks the activation of P2X3 receptors, thereby reducing the hypersensitivity of the cough reflex and decreasing cough frequency. The program is currently in the preclinical stage of development.
