Anocca's p53 R175H A*02 TCR-T is an autologous, T cell receptor (TCR) engineered T cell therapy designed to target the p53 R175H hotspot mutation, which is one of the most frequent oncogenic driver mutations in the TP53 gene. The therapy utilizes high-affinity TCRs that specifically recognize the mutant p53 R175H peptide (HMTEVVRHC) presented by the HLA-A*02:01 MHC class I molecule. To optimize performance within the immunosuppressive tumor microenvironment, the T cells are engineered using non-viral gene editing and incorporate additional functional enhancements, including CD8αβ co-receptors, FAS-41BB switch receptors for improved persistence, and chimeric cytokine receptors. It is currently in Phase 1 development for the treatment of multiple solid tumors harboring the p53 R175H mutation.
