p53MVA vaccine + pembrolizumab is a combination immunotherapy regimen being investigated for the treatment of recurrent, platinum-resistant ovarian, primary peritoneal, and fallopian tube cancers. The p53MVA vaccine is a modified vaccinia Ankara (MVA) viral vector vaccine that expresses the wild-type human tumor protein p53. It is designed to stimulate a T-cell mediated immune response against cancer cells that overexpress or have mutated p53, which is a common occurrence in high-grade serous ovarian cancers. Pembrolizumab is a humanized monoclonal antibody that acts as a checkpoint inhibitor by blocking the programmed cell death protein 1 (PD-1) receptor on T-cells. By blocking PD-1, pembrolizumab prevents the interaction with its ligands (PD-L1 and PD-L2), thereby reversing T-cell suppression and enhancing the anti-tumor immune response. The combination aims to leverage the vaccine's ability to generate p53-specific T-cells and pembrolizumab's ability to maintain their activity within the tumor microenvironment.
