This combination chemotherapy regimen was evaluated in the SWOG Cancer Research Network trial S9914 for the treatment of previously untreated extensive-stage small cell lung cancer (SCLC). The regimen consists of three active cytotoxic agents: **paclitaxel**, a microtubule-stabilizing agent that prevents mitotic spindle breakdown; **carboplatin**, a platinum-based agent that induces DNA cross-linking to inhibit replication; and **topotecan**, a topoisomerase I inhibitor that causes lethal DNA strand breaks. To mitigate the severe myelosuppression associated with this intensive triplet combination, **filgrastim** (granulocyte colony-stimulating factor, G-CSF) is administered as supportive care to stimulate the production of neutrophils. The Phase II study aimed to determine if this multi-targeted approach could improve outcomes in extensive-stage SCLC compared to standard doublet therapies.
