Overview

This combination chemotherapy regimen was evaluated by the SWOG Cancer Research Network in the Phase II trial S9912 for the treatment of optimally-debulked stage III epithelial ovarian, fallopian tube, or primary peritoneal cancer. The regimen consists of intravenous (IV) and intraperitoneal (IP) paclitaxel, IP cisplatin, and IV pegylated liposomal doxorubicin. Paclitaxel is a taxane that works by stabilizing microtubules and preventing cell division. Cisplatin is a platinum-based agent that causes DNA cross-linking, while pegylated liposomal doxorubicin is an anthracycline that inhibits topoisomerase II. Although the trial demonstrated the feasibility of adding liposomal doxorubicin to an intraperitoneal cisplatin-based program, the regimen was associated with severe systemic toxicity and treatment-related mortality, leading to the cessation of its development.
