This is a combination therapy consisting of paclitaxel (a microtubule-stabilizing chemotherapeutic agent), cisplatin (a platinum-based DNA-damaging chemotherapeutic agent), and toripalimab (a monoclonal antibody targeting programmed cell death protein 1, PD-1). Paclitaxel disrupts mitosis by stabilizing microtubules, leading to apoptosis in rapidly dividing cells. Cisplatin forms DNA crosslinks that inhibit DNA replication and transcription, also resulting in cell death. Toripalimab is an immune checkpoint inhibitor that blocks PD-1 on T cells, enhancing the immune system’s ability to recognize and destroy cancer cells. This combination has demonstrated significant improvements in progression-free survival and overall survival compared to chemotherapy alone for patients with advanced or metastatic esophageal squamous cell carcinoma (ESCC) as first-line treatment[1][5][10]. It is also being studied for other cancers such as triple-negative breast cancer and oral squamous cell carcinoma[4][6].
