Overview

This is a combination chemotherapy regimen consisting of three cytotoxic agents: paclitaxel, docetaxel, and ifosfamide. Paclitaxel and docetaxel are both taxane-class antineoplastic agents that promote microtubule assembly while inhibiting their disassembly, thereby disrupting mitosis and inducing apoptosis in rapidly dividing cancer cells. Ifosfamide is an alkylating agent of the oxazaphosphorine group that crosslinks DNA strands, leading to inhibition of DNA replication and cell death. This combination has been explored for use in various advanced or refractory cancers due to potential synergistic effects from targeting different mechanisms within the cell cycle. However, there is limited published evidence on the simultaneous use of all three drugs together as a single regimen; most studies have evaluated two-drug combinations among these agents[1][2][3][4]. The rationale for combining these drugs includes overcoming resistance to individual components (such as taxane resistance) and broadening antitumor activity.
