Overview

Paclitaxel + doxorubicin is a combination chemotherapy regimen consisting of two well-established anticancer agents. Paclitaxel is a microtubule-stabilizing agent that inhibits cell division by blocking the disassembly of microtubules, leading to apoptosis in rapidly dividing cells. Doxorubicin is an anthracycline antibiotic that exerts its cytotoxic effects primarily through inhibition of topoisomerase II, intercalation into DNA, and generation of reactive oxygen species. The combination leverages their distinct mechanisms to enhance antitumor efficacy and reduce the likelihood of drug resistance. This regimen has been widely used in the treatment of advanced and metastatic breast cancer as well as other solid tumors. However, it is associated with significant toxicities including neutropenia, neuropathy, cardiotoxicity (notably from doxorubicin), nausea/vomiting, alopecia, myalgia, and risk for congestive heart failure[1][3]. Combination therapy aims to improve response rates but requires careful monitoring due to overlapping toxicities.
