Paclitaxel + fluorouracil is a combination chemotherapy regimen consisting of two well-established cytotoxic agents used primarily in the treatment of various solid tumors. Paclitaxel is a taxane that stabilizes microtubules and inhibits their depolymerization during cell division, leading to mitotic arrest and apoptosis. Fluorouracil (also known as 5-fluorouracil or 5-FU) is an antimetabolite that inhibits thymidylate synthase and interferes with DNA synthesis by mimicking uracil. The combination exploits synergistic effects by targeting different phases of the cell cycle and mechanisms within cancer cells. This regimen has been investigated for enhanced efficacy in breast cancer and other malignancies such as gastric cancer[1][2][3]. Novel delivery systems such as exosome-based formulations are under investigation to improve oral bioavailability and reduce toxicity[1][2].
