FOLFPTX is an investigational chemotherapy combination regimen consisting of paclitaxel, fluorouracil (5-FU), and calcium levofolinate (leucovorin). It is being evaluated by Fujian Cancer Hospital as a first-line treatment for patients with advanced gastric cancer (AGC) or adenocarcinoma of the esophagogastric junction (AEG). Within this regimen, paclitaxel acts as a microtubule stabilizer that interferes with cell division by preventing the depolymerization of tubulin. Fluorouracil is an antimetabolite that inhibits thymidylate synthase and is incorporated into DNA and RNA, thereby disrupting nucleic acid synthesis. Calcium levofolinate is included as a biochemical modulator to enhance the cytotoxic activity of fluorouracil. This triplet regimen is being compared against standard doublets like mFOLFOX and mFOLFIRI in randomized clinical trials (such as NCT03045770) to determine its relative efficacy and safety profile in the first-line setting.
