Overview

This is a combination chemotherapy regimen consisting of three agents: - **Paclitaxel** is a microtubule inhibitor that stabilizes microtubules and prevents their depolymerization during cell division. This action disrupts mitosis and leads to apoptosis in rapidly dividing cancer cells. - **Gemcitabine** is a nucleoside analog that inhibits DNA synthesis by incorporating into DNA strands and inhibiting ribonucleotide reductase. This results in the inhibition of DNA replication and cell death. - **Liposomal doxorubicin** (specifically pegylated liposomal doxorubicin) is an anthracycline antibiotic encapsulated in a liposome with polyethylene glycol coating. It intercalates into DNA and inhibits topoisomerase II activity, leading to inhibition of DNA replication and generation of free radicals that cause cytotoxicity. The liposomal formulation allows for prolonged circulation time and selective accumulation in tumor tissues due to enhanced permeability[1][2][5][6]. This combination has been studied as a biweekly regimen for advanced solid tumors including ovarian cancer[2]. Each drug works via distinct mechanisms to inhibit tumor growth or induce apoptosis.
