Overview

Paclitaxel + ifosfamide + cisplatin, commonly referred to as the TIP regimen, is a combination chemotherapy protocol primarily used for the treatment of testicular germ cell tumors (GCTs), especially in cases where the disease has relapsed or is refractory to first-line therapy. It is also studied in other cancers such as advanced or recurrent cervical cancer. Paclitaxel acts by stabilizing microtubules and inhibiting their disassembly, thereby blocking cell division. Ifosfamide is an alkylating agent that cross-links DNA strands, leading to apoptosis. Cisplatin forms DNA crosslinks and inhibits DNA synthesis and function, resulting in cell death. The combination exploits different mechanisms of action to maximize cytotoxicity against rapidly dividing tumor cells[1][2][3][4][5].
