This experimental combination regimen is being investigated by the Obstetrics & Gynecology Hospital of Fudan University for the treatment of recurrent ovarian cancer. The regimen integrates standard-of-care chemotherapy (either weekly paclitaxel or nanoparticle albumin-bound paclitaxel) with the PD-1 checkpoint inhibitor pembrolizumab and the beta-3 adrenergic receptor agonist mirabegron. Paclitaxel and nab-paclitaxel are microtubule-stabilizing agents that interfere with mitosis, leading to cell death. Pembrolizumab is a humanized monoclonal antibody that blocks the interaction between PD-1 and its ligands, thereby revitalizing the anti-tumor T-cell response. Mirabegron, traditionally used for overactive bladder, is included in this oncology setting based on research suggesting that beta-3 adrenergic signaling modulation can influence the tumor microenvironment and potentially enhance the efficacy of immunotherapy and chemotherapy.
