Overview

paclitaxel + oxaliplatin is a combination chemotherapy regimen evaluated for use in definitive concurrent chemoradiotherapy (CCRT). Paclitaxel is a taxane-based small molecule that promotes the assembly of microtubules from tubulin dimers and stabilizes microtubules by preventing depolymerization, which results in the inhibition of the normal dynamic reorganization of the microtubule network that is essential for vital interphase and mitotic cellular functions. Oxaliplatin is a platinum-based antineoplastic agent that undergoes non-enzymatic conversion in physiologic solutions to active derivatives which form inter- and intra-strand DNA cross-links. These cross-links inhibit DNA replication and transcription, leading to cytotoxic effects. This specific combination was investigated by Hangzhou Cancer Hospital in a Phase 2 trial for patients with unresectable locally advanced esophageal cancer, where it was administered concurrently with radiotherapy to assess efficacy and safety.
