POF is a triplet chemotherapy regimen consisting of paclitaxel, oxaliplatin, and 5-fluorouracil (5-FU) with leucovorin (calcium levofolinate), specifically investigated by Fujian Cancer Hospital for the treatment of advanced or metastatic gastric adenocarcinoma. The regimen functions through a multi-targeted approach: paclitaxel stabilizes microtubules to inhibit cell division, oxaliplatin induces DNA damage via cross-linking, and 5-fluorouracil inhibits thymidylate synthase to disrupt DNA synthesis, with leucovorin acting as a biochemical modulator to enhance the cytotoxic effect of 5-FU. In clinical studies at Fujian Cancer Hospital, the POF regimen has been evaluated as a first-line therapy, both as a standalone treatment and in combination with the VEGFR-2 tyrosine kinase inhibitor apatinib, demonstrating efficacy in improving outcomes for patients with advanced gastric malignancies.
