Paclitaxel + palbociclib is an investigational combination therapy for cancer, primarily studied in advanced breast cancer. Paclitaxel is a microtubule-stabilizing agent that disrupts mitosis by arresting cells in the M phase of the cell cycle, leading to apoptosis. Palbociclib is a selective cyclin-dependent kinase 4/6 (CDK4/6) inhibitor that blocks cell cycle progression at the G1/S transition by inhibiting phosphorylation of retinoblastoma protein. The rationale for combining these agents lies in their complementary mechanisms: while paclitaxel targets dividing cells during mitosis, CDK4/6 inhibition can synchronize tumor cells or sensitize them to subsequent chemotherapy. Clinical trials have shown that alternating sequential dosing schedules are feasible and may enhance efficacy without additive toxicity, with promising results particularly in Rb-positive advanced breast cancer[1][2][5]. This combination remains under clinical investigation.
