Paclitaxel + pralatrexate is an investigational combination of two chemotherapeutic agents used in oncology research. Paclitaxel is a taxoid mitotic inhibitor that stabilizes microtubules and prevents their depolymerization, thereby inhibiting cell division and inducing apoptosis in cancer cells. Pralatrexate is a synthetic antifolate designed to inhibit dihydrofolate reductase (DHFR), disrupting DNA synthesis and repair by interfering with folic acid metabolism. The combination has shown synergistic or additive antiproliferative effects in preclinical studies and early-phase clinical trials for solid tumors such as non-small cell lung cancer (NSCLC) and lymphomas[2][3]. Both drugs are administered intravenously.
