Paclitaxel + raltitrexed is an investigational combination chemotherapy regimen. Paclitaxel is a taxane that stabilizes microtubules, thereby inhibiting cell division. Raltitrexed is a quinazoline folate analogue that acts as a potent, specific inhibitor of thymidylate synthase (TS), leading to DNA fragmentation and cell death. This combination has been primarily studied by Fudan University Shanghai Cancer Center as a second-line treatment for advanced or metastatic gastric cancer and gastroesophageal junction adenocarcinoma. A Phase II trial (NCT02072317) compared this combination to paclitaxel monotherapy, showing potential improvements in progression-free and overall survival, although the results were not statistically significant.
