Overview

Paclitaxel + sorafenib is a combination therapy consisting of two distinct anticancer agents used together to enhance antitumor efficacy, particularly in various solid tumors. Paclitaxel is a microtubule-stabilizing agent that inhibits cell division by promoting tubulin polymerization and preventing microtubule disassembly, leading to apoptosis in rapidly dividing cancer cells. Sorafenib is a small molecule multikinase inhibitor that targets several protein kinases involved in tumor cell proliferation and angiogenesis, including vascular endothelial growth factor receptor (VEGFR), platelet-derived growth factor receptor (PDGFR), and RAF kinase. The combination has been investigated for synergistic effects against cancer stem cells and advanced malignancies such as melanoma and breast cancer, with studies showing enhanced inhibition of proliferation, self-renewal, colony formation, and differentiation compared to monotherapy[1][2][3][4]. However, clinical trials have shown mixed results regarding overall survival benefit.
