Paclitaxel + thalidomide is an experimental combination therapy consisting of two small molecule drugs with distinct but potentially synergistic mechanisms of action. Paclitaxel is a mitotic inhibitor that stabilizes microtubules, thereby inhibiting cell division and inducing apoptosis in rapidly dividing tumor cells. Thalidomide is an immunomodulatory and anti-angiogenic agent that inhibits the formation of new blood vessels required for tumor growth and modulates immune responses. The combination has been studied preclinically and in early clinical trials for its potential to enhance antitumor efficacy, particularly in highly vascular tumors such as colorectal cancer xenografts and advanced non-small cell lung cancer, by combining cytotoxic effects with inhibition of angiogenesis[1][6][8]. Both agents have established roles as single agents or in other combinations for various cancers.
