Overview

This is a combination regimen of three agents: - **Paclitaxel** is a microtubule-stabilizing small molecule chemotherapeutic that inhibits cell division by promoting tubulin polymerization and preventing microtubule disassembly. - **Trastuzumab** is a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2), used primarily in HER2-positive breast cancer to inhibit proliferation and mediate antibody-dependent cellular cytotoxicity. - **Trebananib** (AMG 386) is a peptibody that neutralizes angiopoietin-1 and angiopoietin-2, preventing their interaction with the Tie2 receptor on endothelial cells. This blocks angiogenesis by inhibiting the Ang/Tie2 signaling axis. The combination has been studied in early-stage and metastatic HER2-positive breast cancer as well as other solid tumors. Clinical trials have shown manageable toxicity profiles for this regimen, with common adverse events including peripheral edema, diarrhea, alopecia, fatigue, nausea, nail disorder, and rash. The combination aims to target tumor cells directly while also disrupting tumor vasculature[1][5][8].
