PaclEVin is a combination chemotherapy regimen consisting of paclitaxel, vinorelbine, and estramustine phosphate, which was investigated by NYU Langone Health in a Phase I/II clinical trial. The regimen utilizes a 'triple tubulin targeting' strategy to combat advanced malignancies, particularly metastatic hormone-refractory prostate cancer. Paclitaxel acts as a microtubule-stabilizing agent, vinorelbine serves as a microtubule-destabilizing vinca alkaloid, and estramustine phosphate is a conjugate of estradiol and nornitrogen mustard that binds to microtubule-associated proteins and the estrogen receptor. This multi-pronged approach is designed to overcome resistance to single-agent microtubule-targeting therapies by disrupting microtubule dynamics through multiple distinct mechanisms.
