Paclitaxel + vintafolide is an investigational combination therapy composed of two agents: - **Paclitaxel** is a well-established chemotherapeutic agent that stabilizes microtubules and inhibits cell division, leading to apoptosis in rapidly dividing cancer cells. - **Vintafolide** (EC145) is a folate receptor-targeted small molecule drug conjugate. It consists of a folic acid ligand linked to desacetylvinblastine hydrazide (DAVLBH), a potent vinca alkaloid cytotoxic agent. Vintafolide selectively binds to the folate receptor alpha, which is overexpressed on certain tumor cells (notably ovarian and some breast cancers), allowing targeted delivery of the cytotoxic payload. The rationale for combining these drugs lies in their non-overlapping toxicity profiles and potential synergistic antitumor effects. Preclinical studies have shown enhanced efficacy when used together compared to either agent alone[2]. Clinical trials have evaluated this combination in advanced triple-negative breast cancer and other solid tumors expressing the folate receptor[1][6][8].
