This chemo-immunotherapy combination regimen is being evaluated by Fudan University for the treatment of locally advanced non-small cell lung cancer (NSCLC). The regimen consists of a nanoparticle formulation of paclitaxel (micelles), the platinum-based alkylating agent cisplatin, the antimetabolite 5-fluorouracil (5-FU), and serplulimab, a humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. In the FANLUNG-2 clinical trial, this combination is utilized as induction therapy in a sandwich model alongside a novel high-low mixed-dose radiotherapy strategy. The approach aims to achieve rapid tumor regression and improve disease control rates while minimizing the risk of radiation-induced pneumonitis by reducing overall lung radiation exposure.
