Paclitaxel nanoparticles (fucoidan-based) is an experimental targeted nanotherapeutic formulation consisting of the cytotoxic agent paclitaxel encapsulated within or conjugated to nanoparticles composed of fucoidan. Fucoidan is a natural sulfated polysaccharide derived from brown algae that exhibits a high affinity for P-selectin (CD62P), a cell adhesion molecule overexpressed on activated vascular endothelial cells and within the tumor microenvironment, particularly following vascular damage. This formulation leverages fucoidan as a bioactive carrier to selectively deliver paclitaxel to tumor sites, thereby increasing intratumoral drug concentration while potentially reducing systemic toxicity. Preclinical studies have demonstrated that when combined with vascular-targeted photodynamic therapy (VTP), these nanoparticles can exploit the VTP-induced inflammatory response and P-selectin expression to achieve sustained drug release and superior tumor control in models of prostate cancer.
