This is a four-drug combination therapy consisting of a nanoparticle-based formulation of paclitaxel (paclitaxel polymeric micelles), the platinum-based chemotherapeutic carboplatin, and two immune checkpoint inhibitors—iparomilimab and tuvonralimab. Paclitaxel acts as an antimitotic agent by stabilizing microtubules, while carboplatin induces DNA crosslinking leading to apoptosis. Iparomilimab is a monoclonal antibody targeting programmed death receptor 1 (PD‑1), and tuvonralimab targets cytotoxic T lymphocyte-associated protein 4 (CTLA‑4). The dual blockade of PD‑1 and CTLA‑4 releases inhibition on T cells, enhancing anti-tumor immunity. This combination is under investigation as neoadjuvant therapy for triple-negative breast cancer[1][2], with related combinations also studied in other solid tumors such as cervical cancer[3][6][7]. The checkpoint inhibitor pair has received conditional approval in China for recurrent or metastatic cervical cancer after platinum-based chemotherapy[6][9].
