**Paclitaxel-spore conjugate (cetuximab-targeted)** is a preclinical EGFR-targeted cytotoxic delivery construct developed by SporeGen for colorectal cancer. It uses killed recombinant *Bacillus subtilis* spores displaying streptavidin as a carrier for paclitaxel and biotinylated cetuximab. Cetuximab directs the spore-associated payload to EGFR-expressing tumor cells, including HT29 colon cancer cells, while paclitaxel stabilizes microtubules through tubulin binding, disrupting mitosis and inhibiting tumor-cell proliferation. In reported in-vitro work, the cetuximab-targeted spore construct increased delivery of paclitaxel to HT29 cells and showed greater antiproliferative activity than water-soluble paclitaxel.
