This combination therapy consists of the CDK4/6 inhibitor palbociclib (PD-0332991), the antimetabolite 5-fluorouracil (5-FU), and the platinum-based chemotherapy agent oxaliplatin. Evaluated in a Phase I clinical trial (NCT01522989) led by Georgetown University, the regimen targets advanced solid tumor malignancies. Palbociclib works by selectively inhibiting cyclin-dependent kinases 4 and 6, which arrests the cell cycle in the G1 phase and potentially sensitizes cancer cells to cytotoxic agents. 5-fluorouracil acts as a thymidylate synthase inhibitor to disrupt DNA synthesis, while oxaliplatin forms DNA cross-links that inhibit replication and transcription. The combination aims to leverage the synergistic effects of targeted cell cycle inhibition and standard cytotoxic chemotherapy to improve outcomes in patients with advanced cancers.
